چکیده

Forty-two piglets on days 3 and 12 of age were used to determine the rate of intestinal absorption of iron by the red cell radioiron method and to evaluate the effect of age on intestinal absorption of iron. s9 Fe-labelled ferric citrate in a dose of 1 2 /aci per kilogram weight was administered with carrier ferric citrate in 5% fructose solution by stomach tube after 1-hr fast. Two levels of carrier dose (8 and 24 mg iron) were assigned. After s 9 Fe dosing, all piglets were daily administered ferric citrate of the same level as carrier dose. Although uptake of s 9 Fe by red cells was markedly lower from the ingested dose than from the intravenous injection, the incorporation into red cells of the ingested s9 Fe reached a plateau 6-days post-dose, suggesting ready incorporation into hemoglobin. The red cell utilization of ingested iron by nursing piglets, dosed with 8 mg iron, ranged broadly from 44 to 85% with mean 59%. Only a small amount of s9 Fe was detected in nonheme iron in the liver at the end of experiment. The uptake of s 9 Fe into red cells for the 24 mg dose was markedly depressed as compared with the 8 mg dose. However, since the amount of S9Fe in nonheme iron in the liver was increased with the elevated dosage, it seems likely that iron absorption from the intestine of the nursing pigs is not largely affected by the elevated dose. Iron depletion prior to initiation of s 9 Fe dosing did not enhance the percentage absorption of iron from the intestine significantly over that of the group receiving 8 mg per os daily. Although hematological parameters did not change with the level of iron dose, nonheme iron in the liver was significantly increased with the elevated dose. Thus, it seems likely that the great physiological iron need for hemoglobin synthesis associated with extremely rapid growth is a major factor responsible for

Calcium does not inhibit iron absorption or alter iron status in infant piglets adapted to a high calcium diet.

The purpose of this study was to investigate whether a dietary calcium:iron ratio similar to that often consumed by premature human infants inhibits iron absorption in infant piglets adapted to a high calcium diet. Male Yorkshire piglets were randomized at 3 to 4 d of age to a high calcium diet (4.67 g/L = HC) or a normal calcium diet (2.0 g/L = NC) and fed for 2 to 2.5 wk. An iron dextran inje...

Dietary hemoglobin rescues young piglets from severe iron deficiency anemia: Duodenal expression profile of genes involved in heme iron absorption

Heme is an efficient source of iron in the diet, and heme preparations are used to prevent and cure iron deficiency anemia in humans and animals. However, the molecular mechanisms responsible for heme absorption remain only partially characterized. Here, we employed young iron-deficient piglets as a convenient animal model to determine the efficacy of oral heme iron supplementation and investig...

Effects of Iron from an Amino Acid Complex on the Iron Status of Neonatal and Suckling Piglets

This experiment was conducted to investigate the effects of iron from an amino acid complex (Availa-Fe) on the iron status of neonatal and suckling piglets. A total of 24 gestating sows (Landrace×Large White) were randomly allocated to three dietary treatments. The control diet contained 80 mg kg Fe from ferrous sulfate heptahydrate (FeSO4⋅7H2O), while the two experimental diets were supplement...

Iron Supplementation in Suckling Piglets: How to Correct Iron Deficiency Anemia without Affecting Plasma Hepcidin Levels

The aim of the study was to establish an optimized protocol of iron dextran administration to pig neonates, which better meets the iron demand for erythropoiesis. Here, we monitored development of red blood cell indices, plasma iron parameters during a 28-day period after birth (till the weaning), following intramuscular administration of different concentrations of iron dextran to suckling pig...

Accumulation and metabolism of iron-dextran by hepatocytes, Kupffer cells and endothelial cells in the neonatal pig liver.

Treatment of newborn pigs with supplemental iron is a common procedure utilized to prevent neonatal anemia. The aim of this study was to investigate the hepatic distribution and intracellular metabolism of iron-dextran, a widely used colloidal-iron-carbohydrate preparation. Piglets were injected intramuscularly with iron-dextran (50 mg Fe/kg body wt) at 1 d of age. Hepatocytes and sinusoidal ce...
